Job Description

Seeking: Scientist (Stem Cell Biologist)

  • Full-time position
  • Competitive compensation (Salary, Benefits & Options)
  • New York City, NY

About Aeolis Therapeutics:

  • Aeolis is a biotech startup building a powerful genomics platform to engineer therapeutics for debilitating genetic diseases with no current treatment options.
  • Our proprietary therapeutic approach is uniquely suited to target hundreds of "undruggable" disease genes.
  • We combine human genetics, single-cell genomics, and state-of-the-art cellular assays to identify safe and effective therapeutic candidates at a fraction of the cost.
  • We are a small, well-funded team looking to expand. In addition to competitive salaries, we offer substantial equity to recruit highly technical scientists who want to help build a company from the beginning.
  • At Aeolis, we value creativity, innovation, and the dedication to making a positive impact on patients’ lives. Get in touch with us if you share these values and believe in pushing the boundaries of computational and experimental biology.

Job Description:

Aeolis is seeking a motivated scientist to join a team using cutting-edge genomics, human genetics, and disease modeling to support the pre-clinical development of therapeutics. We are a fast-paced growing startup looking for candidates who want to have a meaningful impact on the company’s biomedical advances. The ideal candidate will have expertise in experimental and computational approaches to analyze mammalian gene regulation, and will be able to use human iPS cells, primary cells, or organoid/animal models to evaluate therapeutic candidates.

Duties and Responsibilities

  • Investigate transcriptional and post-transcriptional regulation of disease genes and the effects of pre-clinical therapeutic candidates (e.g. small molecules, oligonucleotides).
  • Apply experience with mammalian tissue culture to develop in vitro systems with high translatability to the clinic, including human iPS cells, patient-derived primary cells, or organoids.
  • Develop and implement high-throughput screens (HTS) with CRISPR technologies.
  • Ability to introduce new technologies to improve the performance, throughput, and accuracy of existing assays.
  • Collaborate with human geneticists, computational biologists, molecular biologists, and animal disease-modeling experts to determine the mechanism, safety, and efficacy of therapeutic candidates.
  • Actively engage in prioritizing key milestones and developing appropriate timelines.
  • Provide conceptual input on experimental design and interpretation.
  • Option to coordinate with rare disease foundations.
  • The successful candidate will be able to tackle new problems independently while also working as part of a team with a unified goal.

Required Qualifications

  • Ph.D. in Molecular Biology, Genetics, Developmental Biology, Cell Biology, or related field.
  • 3+ years of academic or industry experience working with mammalian cells.
  • Hands-on experience with iPS cells, primary cells, or organoid systems.
  • Expertise using methods to perturb mammalian gene expression including CRISPRi, CRISPRa, oligonucleotide approaches (siRNAs and ASOs), or gene therapy approaches (AAV systems).
  • Perform, analyze, and interpret next-generation sequencing and genomics assays (RNA-seq, single-cell RNA-seq, PRO-seq, ChIP-seq, or ATAC-seq)
  • Expertise in molecular cloning (InFusion, Gibson), RNA/DNA purification, and qRT-PCR.
  • An excellent scientific track record with published research papers in the field of gene regulation, RNA biology, developmental biology, genomics, translation, or drug discovery.
  • Superb written and oral communication skills, and the organizational abilities to pursue several projects on tight timelines.

Preferred Qualifications

  • Genetic manipulation of mammalian cells using lentivirus transduction, recombinase-mediated integration, artificial chromosomes (BACs or HACs), or CRISPR gene editing.
  • Expertise using flow cytometry and Fluorescence Activated Cell Sorting (FACS).
  • Expertise using immunofluorescence and fluorescence in situ hybridization (FISH) to determine the localization of proteins and nucleic acids.
  • Hands-on experience with mouse or rat disease models.
